Epstein-Barr virus (EBV) is a double stranded DNA herpes virus that can have a profound effect on the immune system. The sero-prevalence of EBV in MS cases is nearly 100% and is associated with the greatest risk for MS among all the infectious agents that have been reviewed. It is not yet clear what determines who will develop MS following EBV infection. A better understanding of the role of EBV infection in MS pathogenesis will greatly improve diagnostic, therapeutic, preventive and reparative strategies.
To address the key knowledge gaps, the PhD candidate will work with a team of leading experts in three key areas: EBV pathogen and host genetics, neurology, and T cell immunity. The proposed aims for the project include 1) evaluate structural variations in EBV pathogen and host genome; 2) single-cell RNA sequencing analysis (scRNAseq) as well as scTCR sequencing on LCL-activated T cells from MS cases and healthy controls
By providing single cell level high-resolution transcriptome data to trace the complex adaptive immune response to EBV infection, we will identify T cell gene signatures specific to MS cases as well as population dynamics of T cells responding to both EBV antigens and self-antigens. This will provide invaluable insights into potential refinements of the T cell immunological therapy that our team has developed, which is now in a phase II clinical trial for MS.

Applicants will be considered for a Research Training Program (RTP) scholarship or Tasmania Graduate Research Scholarship (TGRS) which, if successful, provides:
